Application of real-time PCR for detection and quantification of
Listeria monocytogenes in food.
In this lecture the application of real-time PCR for detection
of Listeria monocytogenes in foods is discussed. Principal as-
pects are discussed: pre-amplification strategies for different
foods in order to remove or minimise the presence of inhibitory
substances present in the foods that consequently it can produce
the subestimation of the bacterial load as well as false nega-
tive results. As well, the application of an adequate control of
the amplification reaction efficiency is discussed. In this
sense, the application of internal amplification controls (IAC)
can allow the assessment and interpretation of the diagnostic
results of the molecular techniques. control. Speed, excellent
detection limit, selectivity, specificity, sensitivity and po-
tential for quantification and automation are among the most im-
portant advantages of real-time PCR for Listeria monocytogenes
diagnosis.
